The gWiz™ Expression Vector has been systematically engineered to produce the highest levels of reporter gene expression in a wide variety of mammalian cells and tissues. The gWiz vector obtains its exceptional gene expression levels from an optimized human cytomegalovirus (CMV) promoter followed by intron A from the CMV immediate-early (IE) gene. In addition, the gWiz vector contains an optimized plasmid backbone that has been extensively altered to achieve abundant plasmid production in E. coli.

CHO-K1 cells were grown in chambered tissue slides and transfected in serum-free conditions with 1 ug of B-gal plasmid (gWiz vector or a commercial plasmid with CMV IE promoter driving B-gal = CMV-1) and 5 ul of GenePORTER® Transfection reagent. CV-1 and NIH 3T3 cells were transfected with 2 ug of plasmid DNA and 10 ul of GenePORTER reagent. After the initial 4 hours of incubation in serum-free media, 10% FBS-containing medium was added to the chambered slides. The cells were then cultured for 48 hours and then stained with X-gal according to standard procedure. Finally cells were examined with a upright microscope (Nikon E-600) equipped with a 20X pan fluor objective and DIC optics.
