Removal of Extracellular Cationic Lipid/DNA Complexes Using CellScrub™ Buffer

Adherent cells were incubated with cationic lipid/DNA complexes at 4°C for 1-2 hours. Several washes removed cell-associated complexes. After washes, cells were lysed, and the DNA was extracted. DNA samples were analyzed by agarose gel electrophoresis. DNA was stained with ethidium bromide. Arrows show plasmid DNA.
Lane 1: Trypsinization and PBS washes.
Lane 2: Washing procedure including the CellScrub™ washing buffer.

Sample Protocol

Culture adherent cells in 6-well plates and transfect with 5 μg of DNA complexed with cationic lipids per well as described.
Remove culture medium and wash cells with PBS (containing CaCl2 and MgCl2).
Treat cells with CellScrub buffer (1-2 ml/well) and incubate at room temperature for 10 to 15 minutes to remove extracellular cationic lipid/DNA complexes.
Wash twice with PBS (without CaCl2 and MgCl2).
