MCF-7 cells were harvested at 90% confluency from a T150 culture flask using a cell scraper, centrifuged and resuspended in 10 mls of complete medium. A. 0.10 mls of suspended cells were transfered to an eppie tube, to which 0.10 mls of PBS was added. The cells were vortexed and incubated @ room temperature for 5 mintues then mounted on a glass slide and photographed. MCF-7 cells are adherent and grow in clumps. They are very difficult to dissociate. B. 0.10 mls of suspended cells were transfered to an eppie tube, to which 0.10 mls of cold FACSmax cell dissociation solution was added. The cells were vortexed and incubated @ room temperature for 5 mintues then mounted on a glass slide and photographed. After treatment with FACSmax a single cell suspension makes for easy and accurate cell counting.
