The new 2EzHRV and 2EzTEV Dual-Tagged Proteases cleave off fusion proteins at sites used in the popular pGEX vectors. Currently, Invitrogen incorporates the TEV site in many Gateway vectors, and is selling the protease as "AcTEV". GE (Amersham) first incorporated HRV 3C protease site in pGEX vectors and call the protease "PreScission protease". The advantages of Genlantis' 2EzHRV and 2EzTEV proteases are: 1. lower price, and 2. the inclusion of two tags on allows for GST or His-tag purification, which means easier purification of the customer's target protein

2EzTEV Dual Tagged Protease

2EzTEV Dual Tagged Protease is a genetically engineered N1a protein that contains the active catalytic fragment of the Tobacco Etch Virus (TEV).The N1a protein is a cysteine protease that recognizes the peptide sequence: Glu-Asn-Leu-Tyr-Phe-Gln-Gly, and cleaves between the Gln-Gly residues. The 2EzTEV Dual Tagged Protease is a 52 kDa molecular biology grade protease that is highly active, specific, and stable at the 4oC working temperature. Furthermore, the 2EzTEV Dual Tagged Protease does not require any special buffer for its activity and can be used in a buffer that is most suitable for the target fusion protein. Best of all, the 2EzTEV Dual Tagged Protease adds great downstream convenience and flexibility since it can be removed by either Ni-chelating or Glutathione(GSH) resins.

2EzTEV Dual Tagged Protease

A 49 kDa GST-fusion protein (C) at 1 mg/ml is incubated with 2EzTEV or competitor TEV Protease (TEV) at a ratio of (1) 1:50, (2) 1:100, (3) 1:200 (w/w) in a buffer of 25 mM Tris-HCl, pH 8.0, 150 mM NaCl, 14 mM B-mecaptoethanol.  Digestion was performed at 4oC for 16 hours. The cleaved products are 27 kDa and 22 kDa.

A 68 kDa GST-fusion protein (C) at 1 mg/ml is incubated with 2EzHRV Protease at a ratio of (1) 1:50, (2) 1:100, (3) 1:200, (4) 1:400 (w/w) in a buffer of 25 mM Tris-HCl, pH8.0, 150 mM NaCl, 14 mM B-mecaptoethanol at 4�C for 16 hours. The cleaved products are 42 kDa and 26 kDa.
